Same here. Gigabyte 450 - I got on a call to the post office, mike just died. Tried another. All settings appear to be correct - no input detected when mike plugged in.

I have been out of this thread for a while. So you guys are saying that an analog microphone plugged into the pink hold on the back of the motherboard doesn't work? I must say I have not done this type of setup for years because I always recommend people use a digital headset with a boom microphone for best results if they want to be heard clearly by the other side. If they won't spring for that I let them use the built in mic of a webcam....probably not much worse in quality then an analog microphone from day of yore. Most of the time people want webcam too when they want a microphone.
I am building another B450M DS3H rig right now. If I can find a old microphone to plug in there I will give it a whirl and let ya' know how I make out

In all the years I have supported people with microphone and speaker problems it has come down to a few basics.....from a desktop computer perspective the audio card built into your motherboard usually has some analog holes on the back. The green hold is intended to drive a pair of speakers or headphones using a stereo mini jack. When i don't hear sound I have to check power on the speaker set. I check physical volume on the speaker set is turned up. I check that the volume slider is up on the desktop (bottom right tray). I check that it is not muted (no slash through the icon). Then I check that the soundcard built in to the desktop is actually the device that is outputting sound (like Realtek Audio).
Sometimes people allow the graphics card sound card to work and that is intended to send sound signals over HDMI when you are connecting a big screen TV and you want the sound to come out of the TV speakers. So check that setting.
If you plug a set or analog stereo mini jack headphones into the bypass jack on the front of your desktop that should disable the mini jack at the rear but that won't work if it was never wired up in the first place. A lot of home system builders miss that part and that can cause a problem too. In that case you can't use the front bypass until you go inside and wire it up.
If you plug headphones in that uses a USB A port then that means the headphones is actually had a digital sound card in it and now you need to switch that speaker output to the new sound card to hear anything and of course there is a new digital microphone too. Now there will be new sound controls on the cord of the headphones that mute and change the volume too.

I have a b450m and the audio doesn't work. The mother board detects the headphones, or the speakers, but the audio works only if I connect my PC to a TV
Im having the exact same issue. My headset is working perfectly fine but my mic is not working it is detected by my pc but when i test my microphone in settings it sounds very muffled and super quiet if i lightly scratch my headset the sound bar goes all the way up but talking barley moves the line please help me i am devastated . Its as if the microphone is in the speakers becuase if i speak into my speackers itmoves more than when i am blowing into the mic.
I have B460M and the mic is not working. I tried several microphones and none of them is working. I tried those on another computer and they all work well. I checked the recording settings and the device manager and all of them are set properly. Can you please suggest what to do next?

I moved that drive to another computer and copied over all data I wanted to keep and did a clean install.
